Abstract

The control of seed-sucking stink bug populations in the soybean production system has been carried out using chemical products. However, insect populations have become resistant to the active ingredients used for their control, resulting in losses of productivity and the use of too many phytosanitary products. Embrapa Soybean, conducts a breeding program for the development of stink bugs resistant and/or tolerant cultivars to these insects. After several years of field trials in the presence of large stink bug populations, today some tolerant cultivars are being released in the market. However, the mediating mechanisms of this interaction are still under investigation. Secondary metabolism, in our hypothesis, may be related to the interaction between tolerant cultivars and this insect. The aim of this study was to determine by untargeted metabolomics from methanolic extracts of green grains of soybean cultivars BRS 543RR (tolerant) and BRS 399RR (susceptible). For extracting, the pods were field collected, when the plants were at R6 stage. Analyzes were performed on and UPLC-QTof-MSE, at negative ionization mode. The raw data were processed by MarkerLynx XS 4.1 (Waters) software and then transferred to multivariate analysis platforms, SIMCA© 14.1 (Umetrics) and MetaboAnalyst 4.0. To identify potential biochemical markers, PCA, HCA and OPLS-DA analyzes were performed. Two compounds belonging to the metabolism of jasmonic acid and triterpene saponins were putatively annotated in BRS 543RR extracts, using MS-DIAL 3.90 and MS FINDER 3.24. Consulting the literature, we observed that these compounds are related to the plant defense to biotic stresses. The information obtained in this research is important to accelerate the selection of genotypes of soybean with resistance and/or tolerance to stink bugs damage. However, further studies should be conducted to obtain a screening protocol based on metabolic differences in soybean genotypes with stink bug resistant characteristics.
